Design Notes

We chose the sequence MGWSCIILFLVATATGVHS, a signal peptide derived from the human immunoglobulin kappa light chain (Igκ). We placed this signal peptide sequence at the N-terminus of our protein constructs. This sequence is a well-known and widely used leader sequence, known for its high efficiency in directing proteins to the ER for proper folding and subsequent targeting and localisation to the membrane. Some other designed igk parts existed, however their sequences were different and without extensive documentation, so we did not know their effectiveness.
